Scientists Decode Genetics Behind Holstein Cow’s Iconic Spots
Researchers have uncovered the genetic basis for the distinctive black and white coat of Holstein-Friesian cows, a breed synonymous with dairy farming worldwide. In a study led by Professor Matt Littlejohn from Te Kunenga ki Pūrehuroa Massey University, scientists identified two specific DNA variants that play a key role in determining the Holstein’s unique coat patterns.
The study reveals that coat patterns are among the earliest traits selected by humans in domesticated animals, reflecting the long-standing influence of farmers on cattle populations. While other breeds, like Herefords and Galloways, have had their genetic markers identified, the genetics underlying Holstein spotting remained a mystery until now.
Using advanced genomic techniques, the research team analyzed the coat patterns of thousands of Holstein cows. They discovered that two genes, KIT and MITF, are responsible for much of the variation in spotting. Professor Littlejohn explained, “These genes are well-known for controlling pigmentation in a range of species, including humans and dogs.”
The variant associated with the KIT gene is particularly intriguing as it operates differently than typical gene variants. Rather than residing within the gene, it regulates it from a separate location on the chromosome. “Think of it like a light switch turning on a light in another room,” Littlejohn added, illustrating the complex interaction of genetic regulation.
The MITF variant further complicates the genetics of coat patterns. It can produce distinct markings in crossbred cattle, leading to variations such as black speckles in dairy crosses or unexpected white face patterns in Hereford crosses. This has practical implications for farmers, especially those who cross Holsteins with Herefords to enhance beef production.
According to Littlejohn, the presence of the MITF variant can result in calves that are less recognizable as Hereford crosses due to irregular spotting. “When ‘splotchy’ faced calves are born, they are often valued less in the market,” he noted. The potential for genetic testing to predict coat patterns could offer farmers a way to maintain the market value of their livestock.
Beyond aesthetic concerns, the research indicates possible animal welfare benefits. Skin pigmentation plays a role in heat absorption and protection from ultraviolet rays. Cattle with darker coats absorb more heat but also benefit from increased UV protection. Understanding how these genes control pigmentation can help farmers select for coat patterns that enhance animal comfort and performance.
“This research not only solves the mystery about how the Holstein gets its spots but also demonstrates how genes can interact to create unique and unexpected patterns,” Littlejohn said. He emphasized the historical significance of these findings, noting that early animal breeders likely selected spotted animals long before the molecular basis of these traits was understood.
The study was supported by the Ministry of Business, Innovation and Employment Endeavor Fund and the Livestock Improvement Corporation. For more detailed insights, the findings are documented in the paper titled “Structural and epistatic regulatory variants cause hallmark white spotting in cattle,” published in March 2024.
This research not only enhances our understanding of Holstein genetics but also opens new avenues for improving cattle breeding practices in the dairy and beef industries.
